缬沙坦对糖尿病大鼠klotho蛋白及氧化应激影响的研究

【摘要】:目的探讨缬沙坦对糖尿病大鼠肾脏klotho蛋白(KL)和氧化应激的影响。方法糖尿病大鼠随机分为糖尿病(DM)组和缬沙坦干预(VA)组,另设对照(NC)组,每组20只。VA组予缬沙坦55mg/(kg·d)灌胃,NC组及DM组予等量生理盐水灌胃,于4周及12周测定大鼠肾功能、血清KL、8-羟基脱氧鸟苷(8-OHdG)及超氧化物歧化酶(SOD)活性,免疫组织化学测定肾脏KL的表达水平。结果与NC组比较,DM组4周和12周时KL含量降低[(4.39±0.35)vs(3.73±0.69)ng/ml,(4.16±0.28)vs(3.30±0.37)ng/ml],SOD活性降低[(1.87±0.84)vs(1.69±0.29)U/ml,(1.79±0.48)vs(1.50±0.26)U/ml],8-OHdG增加[(2.74±0.11)vs(3.32±0.91)ng/ml,(2.86±0.28)vs(3.21±0.25)ng/ml](P0.05);与DM组比较,VA组上述指标亦改善,差异有统计学意义(P0.05)。结论糖尿病大鼠KL的表达降低,缬沙坦可提高糖尿病大鼠肾脏KL的表达,抑制氧化应激反应。
[Abstract]:Objective to investigate the effects of valsartan on renal klotho protein and oxidative stress in diabetic rats. Methods Diabetic rats were randomly divided into DM group and valsartan intervention group. Control group (n = 20) received valsartan 55mg/(kg d (n = 20) and DM group (n = 20) were treated with normal saline. Renal function was measured at 4 and 12 weeks.
