中国大鲵ERα、P450arom基因的克隆及不同发育期4种性别决定相关基因的表达分析

  中国大鲵(Andrias davidianus)是我国特有的有尾两栖动物,也是全球现存两栖类中体型最大的古老物种,属于国家Ⅱ级重点保护野生动物,具有重要的经济、科研和生态价值。然而,近年来由于栖息地的破坏、过度捕杀以及传染病等因素,导致中国大鲵种群数量急剧下降。为了更好地保护这一古老物种,我国科研人员开始了中国大鲵的人工驯养繁殖研究,先后形成了中国大鲵全人工繁育(Captive Breeding)、原生态繁育(Natural Eco-breeding)、仿生态繁育(Imitated Natural Eco-breeding)等3种人工繁殖技术。随着人工繁育技术的突破,中国大鲵人工种群数量剧增。然而,在人工较高水温饲养条件下,幼鲵绝大部分发育成为雄性,造成中国大鲵人工种群性别比例严重失调。为了解决此问题,在幼鲵性腺分化的敏感期,使用一定浓度的外源雌二醇断续暴露40 d,又使得幼鲵已连续4年全部分化为雌性。以上现象表明,中国大鲵性腺分化受到环境水温与外源雌二醇的直接影响。本文在中国大鲵首次克隆了雌激素受体(estrogen receptor alpha, ERa)、细胞色素P450芳香化酶...
